Technicon AutoAnalyzer II

The Adamski [13] procedure is semiautomated. It gives an accuracy of 8.1% and a precision of 8.2% for seawater samples spiked with 3-35 p,g/l organic nitrogen. This procedure is based on the indophenol blue method and was employed using a Technicon AutoAnalyzer II system with the appropriate accessories. Various workers have described automated procedures for determination of low levels of organic nitrogen in seawater [15,16]. 

Organic phosphorus is determined by the difference in phosphorus content of the 1M hydrochloric acid extract measured before and after ignition of the dry sediments at 550°C. In all instances the orthophosphate is determined by using standard Technicon AutoAnalyzer II techniques. Silica does not interfere.

Salinity was obtained using the Harvey method (9) and was checked with a hand-held refractometer (9). Sihcate and phosphate analysis were obtained by a Technicon AutoAnalyzer II using standard procedures described in the EPA Methods Manual (10).

There are also studies that comp)are sp>ectrophotometric and RP-HPLC determinations of iodine concentrations in urine (Bier et al., 1998). In the first one ammonium persulfate was used as an oxidant in the modified ceiic arsenite method. With the use of this sensitive method iodine concentrations can be determined in very small sp>ecitnens (50 pL). A Technicon Autoanalyzer II and a paired-ion-RP HPLC were the basic analytical equipment. The authors found that the precision of this optimized ammonium j ersulfate method yielded inter assay CVs of <10% for urinary iodine concentrations >10 pg/dL The detection limit was 0.0029 pg iodine. There was a high correlation between all three methods (r > 0.94 in any case) and the interpretation of the results was consistent. The authors suggested that the manual ammonium persulfate method could be performed in any routine clinical laboratory for urinary iodine analysis. Another benefit of the described methods is a p>ossibility to process a large number of samples with high accuracy and ininimal technician s time.

Sulphate, chloride (Cl ), nitrate (NO ), and ammonium (NH ) in water were measured with a Technicon Autoanalyzer II system by the methylthymol blue, mercuric thiocyanate, cadmium reduction and sodium nitroprusside methods, respectively. Nutrients were measured in a Technicon AA-II Autoanalyzer every 2 min on a continuous flow of seawater pumped to the instrument. A 2.54-cm inside diameter (ID) nylon hose with the intake at 2.5 m was led on board to a deck-mounted, air-operated double diaphragm pump (Wilden Pump and Engineering Company, Model M-4, Colton, CA).
