Targeted liposomes, binding

Free folic acid is added for competitive inhibition of folate targeted liposome binding to the folate receptor. Consequently, receptor-mediated uptake of these liposomes will also be inhibited (5). 

Target-Sensitive Liposomes. For targeting liposomes to a specific site, antibody-coated liposomes or ligand-coated liposomes are usually used. Target-sensitive liposomes composed of PE and palmitoyl-antibody have been developed these are destabilized on binding to the target and release the contents at the specific site (46)... 

A great number of peptide-based strategies have been developed to promote cell targeting and binding of liposomes. 

Integrin receptor-binding peptides have been used to enhance liposome binding, uptake, and expression (25,47 9). The inclusion of an 0(5pi integrin-targeted peptide into a liposomal complex enhanced transfection efficiency four- to five-fold in Jurkat cells and 10- to 13-fold in TF-1 cells (48). Confocal and electron microscopy revealed that the mechanism of cell entry conferred by RGD peptides on liposomes is predominantly by clathrin-coated endocytosis rather than by phagocytosis (50). 

In the absence of target, no reporter probe-tagged liposomes bind at capture zone... 

To increase liposomal drug accumulation in the desired areas, the use of targeted liposomes with surface-attached ligands capable of recognition and binding to cells of interest has been suggested (see Figure 8.4).
