Standard conditions are 20 hr at 45,000 rev/min using 1 to 5 /ig DNA. Densities are normally calculated with reference to a DNA of known density which is present in the centrifuge cell with the DNA to be analysed. Usually a standard DNA density 10 or 20 mg/ml above or below the sample DNA can be used (e.g. Micrococcus lysodeiktius, p= 1.731 g/ml Escherichia coli or T7 phage, p= 1.710 g/ml Sober 1968). The density is given by... [Pg.464]

Both cyanobacterial thioredoxins were expressed in E. coli. One gram of bacteria yielded approximately 20 mg of thioredoxin. T-1 was able to restore the wild type phenotype in E. coli strains lacking thioredoxin except for the growth of T7 phage (2). T-2 was not effective in the... [Pg.2947]

Infection of cells with DNA phage may lead to the production of a new, phage-spedfic RNA-P. (e.g. T7 phage RNA-P. is a si e-chtiin protein, M, 107,000), or the host RNA-P. may be modfied with the addition of new protein subunits encoded by the phage (e.g. T4 and k phage) in each case the resulting RNA-P. is specific for transcription of phage DNA. [Pg.616]

It has been shown that the endosialidase is the only factor mediating Kl-specificity since the T7 phage lacking the endosialidase gene is unable to infect E. coli K1 strains. However, after enzymatic removal of the K1 capsule by endosialidase, the naked E. coli K1 strain can be infected by T7 [38]. Vice versa, the KIF phage is unable to infect E. coli strains lacking a polysialic acid capsule [108]. [Pg.37]
