Cyclopropanes Simmons-Smith reaction

Even unreactive alkenes undergo the Simmons-Smith reaction (cyclopropanation) in high yield with dibromomethane and this zinc powder. Almost quantitative yields are obtained in the Reformatsky reaction of ethyl a-bromoacetate and this zinc powder. Ethyl a-chloroacetate can also be used if the reaction is conducted at higher temperatures for a longer period.

It is clear that free CH2 is not involved in the Simmons-Smith reaction, but there is substantial evidence to indicate that caibenes aie formed as intermediates in certain other reactions that convert alkenes to cyclopropanes. The most studied exanples of these reactions involve dichlorocaibene and dibromocaibene. 

Without question, the most powerful method for cyclopropane formation by methylene transfer is the well-known Simmons-Smith reaction [6]. In 1958, Simmons and Smith reported that the action of a zinc-copper couple on diiodomethane generates a species that can transform a wide variety of alkenes into the corresponding cyclopropanes (Scheme 3.3) [7].

A carbene, R2C , is a neutral molecule containing a divalent carbon with only six valence electrons. Carbenes are highly reactive toward alkenes, adding to give cyclopropanes. Nonlialogenated cyclopropanes are best prepared by treatment of the alkene with CH212 and zinc-copper, a process called the Simmons-Smith reaction.

The Simmons-Smith reaction has been used as the basis of a method for the indirect a methylation of a ketone. Chemo- and stereoselective reduction of (56) to (55) is achieved In highest yield by sodium borohydride in ethanol. The isolated ketone is reduced more rapidly than the enone and (55) is the equatorial alcohol. Protection moves the double bond out of conjugation and even the distant OH group in (54) successfully controls the stereochemistry of the Simmons-Smith reaction. Addition of TCS 14 to CH2l2/Zn, which contains up to 0.04 mol% of lead impurity, improves the Simmons-Smith reaction of olefins such as cyclooctene to give up to 96% of the cyclopropane 2103 [36] (Scheme 13.12).

Two examples of substituted cyclopropane formation are shown below. This reactive zinc should dramatically increase the utility of the Simmons-Smith reaction. 

Bernardi and co-workers investigated the mechanism of the Simmons-Smith reaction theoretically at the DFT (B3LYP) level of theory, using ClCH2ZnCl as a model system.95 Of the two available reaction channels (addition and insertion), the former process was found to have the lower activation energy (107.7 vs. 150.7 kj moP1), and this result correlates well with the exclusive cyclopropanation over the competing insertion reaction. 

Furthermore, cyclopropane structures have often served as intermediates in organic synthesis. For these reasons, olefin cyclopropanation has proved to be a useful tool for synthetic organic chemists. This has led to the development of several methods for cyclopropanation reactions,91 including the metal-catalyzed reactions of diazo compounds with olefins, as well as the Simmons-Smith reaction. 

Taking 1,2-disubstituted cyclopropane as an example, retro synthesis analysis shows that there are three possible ways to disconnect the three-membered ring—a, b, and c as shown in Figure 5-11. Route a involves the addition of methylene across a double bond, and this is often a stereospecific conversion or Simmons-Smith reaction.92 One can clearly see that route b or c will encounter the issue of cis/trans-product formation. 

Among methods of preparing optically active cyclopropane compounds, the Simmons-Smith reaction, first reported in 1958, is of significance. This reaction refers to the cyclopropanation of alkene with a reagent prepared in situ from a zinc-copper alloy and diiodomethane. The reaction is stereospecific with respect to the geometry of the alkene and is generally free from side reactions in contrast to reactions involving free carbenes. 

The Simmons-Smith reaction " and its variants are widely used for the stereospecific synthesis of cyclopropane compounds. The methodology involves the use of copper-treated zinc metal (the zinc-copper couple) with diiodomethane to add methylene to a carbon-carbon double bond. Alternative use of diazomethane in catalytic reactions does not offer the same synthetic advantages and is usually avoided because of safety considerations. As significant as is the Simmons-Smith reaction for cyclopropane formation, its employment for organic synthesis was markedly advanced by the discovery that allylic and homoallylic hydroxyl groups accelerate and exert stereochemical control over cyclopropanation of alkenes (e.g, Eq. 21), and this acceleration has been explained by a transition state model... 

The Simmons-Smith-type cyclopropanation of olefins is one of the most well-known reactions of carbenes and carbenoids. However, cyclopropanation of simple olefins with magnesium carbenoids is usually very difficult and only cyclopropanation of allylic alcohols was reported. Thus, treatment of allylic alcohols (23) in CH2CI2 at —70°C with i -PrMgCl and diiodomethane for 48 to 60 h afforded cyclopropanes in up to 82% yield as a mixture of syn- and and-isomers. In this reaction, 5yn-isomers were mainly or exclusively obtained (synianti = 5 1-400 1) (equation 10). 

The presence of an excellent recent review in this area [7] will curtail discussion to three papers. Taguchi et al. [360] showed that protected /Lfluoroallylic alcohols underwent Simmons-Smith reactions more slowly than the non-fluori-nated congeners, but still afforded useful yields of cyclopropane products (Eq. 140) with an excess of the methylene transfer reagent. 

Asymmetric cyclopropanation of olefins can also be achieved by the Simmons-Smith reaction (231). Reaction of ( )-cinnamyl alcohol and the diiodomethane-diethylzinc mixed reagent in the presence of a small amount of a chiral sulfonamide gives the cyclopropylcarbinol in up to 75% ee (Scheme 97) (232a).
