Oftentimes, the objective of preparative flash separations may be different firom the objective of analytical separations. Speed and sensitivity may be less important than product purity in preparative chromatography. Preparative columns may operate at flow rates lower than in analytic columns with gradient profiles altered in order to compensate for the less efficient mass transfer of larger adsorbent particles. 

There are four types of position-sensitive detectors Wire detectors (one- and two-dimensional), video detectors. CCD (charge coupled device chip), and image-plate detectors. The wire detector is bused on the proportional counter principle voltage pulses produced by quanta incident at different points along the wire take different times to reach its two ends. The travel times are derived from the profile of the sum of the two pulses. In order to detect the intensity change of one mode in the presence of many others, the laser output has to be dispersed by a monochromator or an interferometer. The absorbing molecules may have many absorption lines within the broad-band gain profile of a multimode dye laser. Those laser modes which overlap with absorption lines are attenuated or even completely quenched. This results in "spectral holes" in the output spectrum of the laser and allows the sensitive simultaneous recording of the whole absorption spectrum within the laser bandwidth, if the laser output is photographically recorded behind a spectrograph. ATKINSON et al. [8.17] demonstrated this technique by monitoring with a flash lamp-pumped dye laser the time-dependent concentration of the short-lived radicals NH2 and HCO which had been formed by flash photolysis of NH. ...
