Reserpine dosage

Second and subsequent weeks Increase dosage to 50 mg 4 times daily. Maintenance Adjust dosage to lowest effective level. Twice daily dosage may be adequate. In a few resistant patients, up to 300 mg/day may be required for a significant antihypertensive effect. In such cases, consider a lower dosage of hydralazine combined with a thiazide or reserpine or a beta-blocker. However, when combining therapy, individual titration is essential to ensure the lowest possible therapeutic dose of each drug. 

A non-aqueous titration method is described for the determination of some alkaloids including reserpine and their dosage forms using 0.005M chloranilic acid solution in l,it-dioxane as the titrant. The end point is determined by measuring the change in absorbance of the sample at 535 nm. Quantitative recoveries with good reproducibility are reported for reserpine and other alkaloids (38). 

A third method is described for the quantitative determination of chlorthalidone in pharmaceutical dosage forms containing reserpine (Ul). 

Reserpine [0.1 mg/kg intramuscularly (i.m.)] or yohimbine (0.01 mg/kg i.m.) induces significant impairments in the monkey s ability to perform the delayed response task. HA at a dosage of 0.01 mg/kg i.m. for the yohimbine-treated monkeys markedly improved the memory impairments. The effects exhibited an inverted U-shaped dose-response pattern. The data suggest that HA may improve working memory via an adrenergic mechanism. ... 

The administration of berberine (1-4 mg/kg, i.v.) to rabbits was observed to significantly and dose-dependently elevate the ventricular fibrillation threshold (VFT) induced by electrical stimulation. Neither reserpine nor vagotomy exerted any influence on the increase of VFT with berberine, which implies that the anti-arrhythmic action is most likely due to a direct activity on the myocardium, without involvement of the autonomic nervous system. The pronounced effect of the alkaloid on action potentials in the rabbit myocardium in vivo resulted in the reduction in the maximal velocity of depolarization (V ). At higher dosage, berberine produced a prolongation in action potential duration at both 90 and 100% repolarization, in addition to a more potent decrease in V. These results may thus contribute to the effectiveness of berberine against myocardial arrhythmias [256]. 

Different distribution characteristics were also observed39 for dopamine (5) and its tris(trimethylsilyl) and tetrakis(trimethylsilyl) derivatives 6 and 7, respectively. In contrast to 5, for which the blood-brain barrier is nearly impermeable, the much more lipophilic silyl derivatives 6 and 7 were found to exhibit central effects when given intraperitoneally to rats. At a dosage of 100 mg per kg, they influence positively the rigid akinetic syndrom, which was induced by reserpine. These results may be explained in terms of a pro-drug behavior of 6 and 7. The phenothiazine derivatives are hypothermic, and the extent of hypothermia depends on the dosage and the environmental temperature. Substances that reduce the concentrations of norepinephrine (reserpine) or block its receptor site (chlorpromazine) are hypothermic, whereas substances that increase the release of norepinephrine (amphetamine) are hyperthermic. 

A matched pair study of the in-patient population of a psychiatric hospital showed a relatively lower incidence of breast cancer in the married female and the married woman with children. As for reserpine use, checked for dosage and as far as possible for duration of use, no significant difference was found between the breast cancer group and controls (79 ).
