Reference cell lines requirements

The selection of the best cell line should consider antibody productivity, as well as cell growth rate, although these parameters frequently follow opposite trends (Wurm, 2004). For the production of large amounts of mAb, it is fundamental that the productivity of the selected cell line is high. Otherwise, larger reaction volumes will be required, and the cost of purification will be increased. A reference value for specific protein productivity is 20 pg/cell per day. 

Finally, proliferation can be measured by determining the proportion of cells able to form colonies. This is sometimes referred to as a reproductive assay, but again is difficult to perform with fish cell lines. In addition to their slow growth, many fish cell lines proliferate poorly at the low density required for the assay and/or form diffuse colonies, which are troublesome to count. Colony formation has been studied with a few fish cell lines after their exposure to ecotoxicants, with results similar to those from other cytotoxic endpoints17,189.
