Abstract Quantitative receptor autoradiography methods have heen widely used over the last three decades to study the distribution and physiological role of a receptor in various tissues. This review provides an overview of in vivo and in vitro receptor autoradiography methods and their advantages as well as disadvantages in the study of receptors in the central nervous system. Comparison with immuno-histochemical and in situ hybridization methods is also highlighted in relation to the study of a given receptor in the nervous sytem. [Pg.276]

Receptor autoradiography and immunohistochemistry are used to demonstrate the final location of receptor proteins. In the nervous system, these receptor proteins are synthesized within soma, but are generally transported to dendrites or axons, distant from cell bodies where they are originated. Thus, demonstration of a receptor population by receptor autoradiography or immunohistochemistry is not sufficient to directly discriminate which perikarya synthesize the receptors (Kuhar et al, 1986 Quirion et al., 1993 Chabot et al., 1999). However, receptor autoradiography and in vitro studies have suggested that H3 receptors are located on other aminergic neurons in the brain. Since amines such as serotonin and catecholamines are involved in the regulation of pituitary hormone secretion, it is obvious that an action of the H3 receptor compounds may be exerted via these H3 heteroreceptors. Only few studies have evaluated this heteroreceptor action. It has been excluded that the effect of the H3 receptor agonists is due to an effect on H3 receptors located on serotonergic neurons, while an effect on catecholaminergic neurons has yet not been excluded. [Pg.55]
