Protein A chromatography

Protein A chromatography yields inununoglobulins in very concentrated form. Therefore, the absorbance of the solution should not be measured directly. Dilute the sample into PBS, and take areading using PBS as a blank. The optical density of the diluted sample should not be above 1-1.5 to fall within the range of linearity.

Purification of Antibodies Humanized antibodies (hAB-1 and hAB-2) were purified by the technique of Protein-A chromatography, followed by Phenyl Sepharose and Ion-exchange chromatographies. The murine antibody (pMAB) was purified by Protein-A chromatography. 

Cysteine to Serine Conversion The heavy chain CDR-1 residue of hAB-1 was converted to a serine residue by standard in vitro mutagenesis techniques. The mutation was confirmed by sequencing the entire cDNA. The modified CS-hAB-1 was purified by Protein-A chromatography. 

Our method of choice to quantify recombinant antibodies produced in PER.C6 cells is that of analytical protein A chromatography (APAC). This method is automated, rapid (total run time of 3.8 min), precise (standard deviation <5%), and sensitive (quantitation limit 25pgmL ), and generally outperforms ELISA methods, except for sensitivity.
