Preparation of electroporation competent E. coli TGI strain cells

Using sterile techniques, streak E coH TCl strain cells from a glycerol stock onto a minimal medium plate. Grow at 37 °C for 24-36 h.  [Pg.53]

Inoculate a single colony from the overnight E coh TGI strain, streaked on minimal medium plate, into 10 ml of 2 x YT medium and grow overnight at 37 C in a [Pg.53]

Inoculate 1 litre of pre-warmed 2 x YT medium with the 10 ml overnight culture of E. coli TGI strain cells and incubate at 37 C with shaking at 250 r.p.m. for 2-3 h until an ODgoj reading of 0.5-0.7 is reached. For 1 litre culture, use two 5 litre [Pg.53]

Pour the supernatant ofT and resuspend the cells in l litre of ice-cold sterilized Milli-Q water taking care not to lyse them. Centrifuge as in step 4. [Pg.53]
