Dipyridamole is a potent coronary vasodilator. Despite the fact that it causes a substantial increase in CBF, its value for the acute relief or prevention of angina pectoris has been seriously questioned. Considerable effort has been expended in the study of this drug. Deutllcke reports dipyridamole to inhibit adenosine deaminase which results in an accumulation of adenosine. Since adenosine is a potent vasodilator, there is an increase in CBF. Emmons found dipyridamole to inhibit platelet agglutination and proposed that dipyridamole prevents the uptake of adenosine by red blood cells, thereby delaying its deamination. Stafford feels that dipyridamole inhibition of adenosine deaminase is unlikely, as high concentrations are necessary to produce inhibition of deaminase from intestinal mucosa. Much lower concentrations of dipyridamole potentiate the action of adenosine on myocardial tissue. This test requires platelet-rich plasma from a patient recovering from a drug-induced platelet lysis. Various concentrations of the suspected drug have to be used and the mixture incubated in the presence of the patient s serum. Platelet agglutination is best determined on a siliconized slide, using a phase contrast microscope. An indirect method using platelets from a normal donor can also be adapted. It is, however, less sensitive and reliable (Dausset et al. 1964).

Hamilton and Sheets (1978) observed a case of thrombocytopenic purpura after sulfisoxazole which could be demonstrated by the provocation test and by the presence of the serum factors which caused a platelet agglutination in the presence of sulfisoxazole. Antibody investigations have not been carried out routinely in sulfonamide allergies.

The synthesis of 4-trifluoroacetamidophenyl 3,4,6-tri-0-acetyl-2-0-(a-D-glucopyranosyl)-iS-D-galactopyranoside has been reported. After deacetylation, this derivatized disaccharide, which is of interest in blood platelet agglutination studies, is suitable for linking to proteins.
