5-Phenyl-pentanoic acid

Optically pure a-amino acids Ccui be prepared by this route. For example, pure cyclo(L-Aba-L-Lys(e-Ac)) obtained from cyclo (AAba-L-Lys(e-Ac)) was hydrolyzed by 6 M HCl to give pure L-Aba (L-2-aminobutanoic acid) ( 3). Pure L-App (L-2-amino-5-phenyl-pentanoic acid) was prepaured from cyclo(AApp-L-Ala) and used for the synthesis of AM-toxin II ( ). 2H2-D-Phe was prepared from cyclo(APhe-D-Lys(e-Ac)) and deuterium at 0 C, and synthesis of (2H2-D-Phe ]-gramicidin S (cyclic decapeptide) for NMR investigation is under study. 

Benzyl cyanide is first reacted with 2-butylbromide in the presence of sodium amide to give 2-phenyl-3-methylvaleronitrile which is hydrolyzed by sulfuric acid to give 3-methyl-2-phenyl-pentanoic acid. 24 g of 2-phenyl-3-methyl-pentanoic acid are heated for one hour at 175° to 185°C with 30 g of 2-diethylaminoethanol and 0.5 g of sodium methylate. The excess diethyl-aminoethanol is removed in vacuo, the residue is dissolved in 300 cc of 2 N-acetic acid, the acid solution is shaken with ether and made alkaline with concentrated potassium carbonate solution and ice. The ether solution Is washed with water, dried with sodium sulfate and evaporated. The residue is distilled under high vacuum, yielding 20 to 21 g of the basic ester (60% of the theoretical) is obtained, the ester boiling at 98° to 100°C at a pressure of 0.03 mm. The hydrochloride of the ester melts at 112° to 113°C and the methobromide at 100° to 101°C.

ZK 216348 another nonsteroidal structure (Figure 9.4) has been made and characterized by scientists from Bayer Schering Pharma AG [71]. The compound is a pentanoic acid 4-methyl-l-oxo-lH-2,3-benzoxazinamide bearing a trifluoromethyl-and a hydroxyl group at the a-carbon and an additional methyl and phenyl group at the (D-end resulting in a compound with two quaternary carbons.

It is interesting to note that in fermented products the same alcohols always form, even if different sources of nitrogen, but not singular aminoacids, are available. Ketoacids can also be directly reduced to hydroxyacids such as lactic, 2-hydroxy-3-methyl butyric, 2-hydroxy-3-methyl pentanoic, 2-hydroxy-4-methyl pentanoic, 2-hydroxy-3-phenyl propionic and 2-hydroxy glutaric acids. The corresponding ethyl esters can dramatically influence the wine aroma.
