Penicillins production from Penicillium

Initially, enzymes were only used to liberate fhe 6-aminopenicillanic acid (6-APA) nucleus from the fermentation product penicillin G/V from Penicillium chrysogenum using Penicillin G acylase (Scheme 4.6A), and 7-aminocephalospora-... [Pg.101]

The structure of this antibiotic was elucidated in 1945. The most notable feature of this structure is the four-membered p-lactam ring. This unusual feature is key to the antibacterial function of penicillin, as noted earlier (p. 222), Three steps were crucial to fully capitalize on Fleming s discovery. First, an industrial process was developed for the production of penicillin from Penicillium mold on a large scale. Second, penicillin and penicillin derivatives... [Pg.1009]

Characterization of Biosynthetic Pathways. The productivity of Penicillium chrysogenum could have been augmented impressively during the last decades Penicillin titers have been increased by a factor of 50,000 from a very few milligrams/ Liter in the 1940s to more than several lOg/L by now and also some several lOg/L cephalosporin are currently gained. [Pg.13]

A few reports have appeared in the literature on the use of living immobilized whole cells for producing traditional fermentation products (3,12). In this paper, our work on the immobilization of Penicillium chrysogenum ATCC 12690 in polyacrylamide gel, collagen membrane, and calcium alginate and the production of penicillin G from glucose by the immobilized mycelium are described. [Pg.59]

With increasing amounts of AFA in Hansenula polymorpha cultivation medium, kLa passes a maximum at 1.0 %o (w/w) Desmophen. However, gas holdup attains the maximum at 10%o (w/w) Desmophen [41]. The surface tension diminishes and the surface viscosity increases with increasing antifoam concentration in distilled water and in BSA solution. Foaminess 2 drops from 200 to 1 s already at low concentrations of Desmophen (< 1 mgl" ). In a cultivation media of Hansenula polymorpha and E. coU, the surface tension and surface viscosity remain constant during the entire cultivation time in the presence of Desmophen [22]. As already pointed out, the maximum of foaminess which arises at the transition from the growth phase to the production phase during the production of penicillin V by Penicillium chrysogenum can be eUminated by adding Desmophen to the cultivation medium. The foaminess is reduced from 400 to 100 s [26]. [Pg.202]

Penicillium cells immobilized by copolymerization with acrylamide, by co-casting with collagen followed by glutaraldehyde-cross-Iinking, and by coprecipitation with calcium alginate have been used for the production of penicillin G from D-glucose. ... [Pg.671]

Penicillins. Since the discovery of penicillin in 1928 as an antibacterial elaborated by a mold, Penicillium notatum the global search for better antibiotic-producing organism species, radiation-induced mutation, and culture-media modifications have been used to maximize production of the compound. These efforts have resulted in the discovery of a variety of natural penicillins differing in side chains from the basic molecule, 6-aminopenici11anic acid [551-16-6], These chemical variations have produced an assortment of dmgs having diverse pharmacokinetic and antibacterial characteristics (see Antibiotics, P-lactams). [Pg.403]

Penicillins, like most antibiotics, are secondary products whose synthesis is not directly linked to growth. The enzymes that produce secondary products are normally repressed or inhibited under conditions which favour rapid growth. In the early work on penicillin, Penicillium rwtatum was grown as a floating mycelium on about 2 cm depth of liquid medium. The mycelium absorbed nutrients from the medium and penicillin was excreted into the medium. The mycelium and spent medium are readily separated. [Pg.156]

This type of filter can be used for removal of Penicillium and Streptomyces mycelia in the production of penicillin and streptomycin, respectively.5 In these processes the rotary drum filter is used with a pre-coated cloth filter with filter aid the filter cake is removed by a knife blade which scrapes the cake from the rotating drum. [Pg.174]
