The same transformation has been employed by Lei Tao and coworkers for the preparation of a fluorescent protein-reactive poly (ethylene glycol) (PEG) [64]. Synthesis of the latter consists of the aminolysis of dansylthiolactone 9 using commercial monofunctional PEG-NH2 ( 5 kDa) as a nucleophile in the presence of 2,2 -dipyridyl disulfide (Scheme 19). Once prepared, this PEGylation agent can subsequently react with bovine serum albumin (BSA) as a model therapeutic protein to form a fluorescent PEGylated protein as a model of sophisticated theranostic combination, with almost complete preservation of structural integrity and thus bioactivity of the protein. [Pg.125]

Interferon alfa is an antiviral agent that is effective in suppressing hepatitis B viral replication. Interferon alfa-2b and pegylated interferon alfa-2a are the only interferon therapies approved for the treatment of chronic hepatitis B. [Pg.353]

The availability of endogenous enzymes and their variants has allowed the use of enzymes as replacement therapy and as therapeutic agents. With continued refinement in recombinant protein technology, the cost of human recombinant proteins may become more affordable. As more novel human recombinant enzymes are developed, a wide variety of medical disorders will be amenable to enzyme therapies. Additional molecular engineering such as pegylation or creation and modification of... [Pg.254]

Pegylated a-IFN is made by attaching polyethylene glycol (PEG) to the a-IFN. PEG is a large water-soluble molecule that decreases the clearance of a-IFN and also increases the duration of its activity. This modified cytokine is used to treat chronic hepatitis C. However, it is rarely used as a single therapeutic agent for hepatitis C because of its low response rate. [Pg.45]

Efficacy appears to be superior to therapy with nonpegylated interferons in controlled clinical trials, particularly as regards the proportion of patients with sustained virologic responses. As with the nonpegylated interferon alfa agents, combination therapy of the pegylated interferon alfa compounds with ribavirin is more effective than monotherapy. [Pg.1149]
