And so the skillful selection, introduction, and removal of a total of 12 different protective groups have played a major role in the successful total synthesis of paly toxin carboxylic acid (Figure 1,2). 

The C.lOO-C.lOl diol group, protected as an acetonide, was stable to the Wittig reaction used to form the cis double bond at C.98-C.99 and to all of the conditions used in the buildup of segment C.99-C.115 to fully protected paly-toxin carboxylic acid (Figure 1,1). 

Photochemical equilibration of the 3 2 stereoisomeric mixture of N-acyl vinylogous ureas in DMF by irradiation at 300 nm in a Ray-onet reactor equipped with a stannous chloride filter solution at 37 °C for 4 h leads to a 6 1 mixture of trans-Aa b and c .v-Aa b paly-toxins. The total synthesis of palytoxin (1) is now complete. 

In spite of the experimental evidence, the role of the Na-K-ATPase in the formation of the palytoxin channel has not been clarified in detail. The recent advances in the understanding of the Na-K-ATPase function have recently allowed Artigas and Gadsby (2003) to postulate that palytoxin could disrapt the strict coupling between the pump s inner and outer gates, allowing them to both be open. Thus, it is not clear whether the paly toxin-sensitive chaimel is located within the enzyme and the step(s) in the normal functioning of the pump(s) that are affected by palytoxin remain to be elucidated. 

In primary cultures of cerebellar granule cells palytoxin produced a concentration-dependent depolarization of the cell membrane, as assessed with the membrane potential sensitive probe bis-oxonol (Fig. 6.1). Significant differences between control and paly toxin-treated cells were found at palytoxin concentrations equal or higher than 5 nM.

Notable among these are the epochal conquests of vitamin Bi2 by Woodward and Eschenmoser [3] and paly toxin by Kishi [4]. These signal achievements, and many others, have caused many observers, at least in the United States, to question the value of continued research in the area of synthesis, both total synthesis and methods development.
