P-Mannosidase

There is no information so far available about mannan degrading enzymes of alkaliphiles. In this paper, a P-mannan degrading alkaliphile Bacillus sp. AM-001) with high productivities of cell-associated P-mannosidase and three extracellular P-... [Pg.52]

Bacillus sp. AM-001 was cultivated aerobically under various conditions, and activities of P-mannanase in the culture broth and P-mannosidase extracted from the cells treated with 0.1% Triton X-100 were monitored. Both enzymes were formed when the bacterium was grown under alkaline conditions in the presence of optimal concentrations of 0.5% Na2C03 or 0.5-1.0% NaHCOa. Various carbohydrates were also tested and the best carbohydrate for enzyme production was konjak powder (1%... [Pg.53]

The molecular weight (94,000) was smaller than that of other microbial P-mannosidases such as Aspergillus o/jCTe( 120,000-135,000) and Tremella fuciformis (160,000-200,000). The final product from the oligosaccharides with the enzyme was mainly D-mannose and the enzymatic activity was not inhibited by D-mannose or mannose-derivatives such as D-mannosamine, D-mannonic acid and D-mannitol. [Pg.55]

As described in the previous section, this strain produced significant amounts of three extracellular P-mannanases and a cell-associated P-mannosidase. The three P-mannanases differed in several enzymatic properties including optimum pH for enzyme action, optimum temperature, pH stability, thermal stability, isoelectric point and molecular weight. To elucidate the genetic basis for production of multiple forms,... [Pg.55]

In their search for conformationally biased mimics of mannopyranosylamines, A. Vasella and co-workers planned to synthesize compounds that would inhibit p-mannosidases. In order to construct the bicyclo[3.1.0]hexane framework, a five-membered 0-silylated A/,A/-dimethyl-amino alcohol was synthesized. Oxidation of the tertiary amine with mCPBA yielded 83% of the A/-oxide, which was subsequently subjected to the Cope elimination to give 69% of the desired benzyl enol ether. Cyclopropanation of this enol ether gave rise to the highly functionalized bicyclic skeleton. [Pg.97]
