Genetic basis

It s really the ultimate in biological control. That is the determination of structure and function of the next generation by the previous generation. This is the effect of genetic inheritability. 

Genetic material has been called the blueprint of life. It is the material that stores accumulated information about the complexity determined from generations upon generations of adaptations and modifications to environmental pressures.

The VFe protein also has the equivalent of P-cluster pairs which have similar properties to those found in the MoFe protein (159). No information is available on whether P-cluster pairs exist in the FeFe protein, but because of the relatively high sequence identity and the similar genetic basis of its biosynthesis, the occurrence seems highly likely. The catalytic role assigned to the P-cluster pair involves accepting electrons from the Fe protein for storage and future deUvery to the substrate via the FeMo-cofactor centers. As of this writing (ca early 1995), this role has yet to be proved.

The last four disorders cause hemolytic anemias, as do a number of the other disorders listed. Most of the above conditions are discussed in other chapters of this text. MiM numbers apply only to disorders with a genetic basis.

The genetic basis of at least some aspects of the proliferation response has recently been uncovered. Work with Arahidopsis mutants has shown that a nitrate-inducible gene (ANRI) encodes a member of the MADS box family of transcription factors (87). Repression of this gene resulted in plants that no longer responded to nitrate-rich patches. 

The genetic basis for the more common late-onset AD appears more complex. Genetic susceptibility is more sporadic and it may be more dependent on environmental factors.9 The apolipoprotein E (apo E) gene on chromosome 19 has been identified as a strong risk factor for late-onset AD. There are three variants of apo E however, carriers of two or more of the apo E4 allele have an earlier onset of AD (approximately 6 years earlier) compared with non-carriers.9 Only 50% of AD patients have the apo E4 allele, thus indicating it is only a susceptibility marker. 

Results of family and twin studies suggest a genetic basis for bipolar disorder.4 The lifetime risk of bipolar disorder in relatives of a bipolar patient is 40% to 70% for a monozygotic twin and 5% to 10% for another first-degree relative.
