it is important to realize that just because there is no change in the spectrum does not necessarily mean no change in the overall secondary structure of the protein. For example, a polypeptide chain scission may take place inducing a break in a disordered region, leaving individual separate portions of secondary structure intact. 

An advantage of this technique is that an external probe molecule is not required and the absorption of the protein groupings reflect their genuine environments. Techniques such as fluorescence are limited, to a certain extent, by the perturbation which added probes may induce.
