When a mixture of methyl phenyl sulfide (69a) (1 g, 8.1 mmol), 30% H2O2 (1.84 g, 16.2 mmol), and water (10 ml) was stirred at room temperature for 24 h, rac-lOa was produced (Scheme 11). To the water suspension medium of rac-70a was added 10c (2 g, 4 mmol), and the mixture was stirred for 15 h to give a 1 1 inclusion complex of 10c with (+)-70a. Heating the filtered complex in vacuo gave (+)-70a of 57% ee (0.45 g, 82% yield). The one-pot dynamic kinetic resolution (DKR) of ( )-l-phenylethanol lipase esterification in the presence of zeolite beta followed by saponification leads to (R)-l phenylethanol in 70 % isolated yield at a multi-gram scale. The DKR consists of two parallel reactions kinetic resolution by transesterification with an immobilized biocatalyst (lipase B from Candida antarctica) and in situ racemization over a zeolite beta (Si/Al = 150). With vinyl octanoate as the acyl donor, the desired ester of (R)-l-phenylethanol was obtained with a yield of 80 % and an ee of 98 %. The chiral secondary alcohol can be regenerated from the ester without loss of optical purity. The advantages of this method are that it uses a single liquid phase and both catalysts are solids which can be easily removed by filtration. This makes the method suitable for scale-up. The examples given here describe the multi-gram synthesis of (R)-l-phenylethyl octanoate and the hydrolysis of the ester to obtain pure (R)-l-phenylethanol. [Pg.133]

To overcome this issue Kureshy et al. [55, 56] reported dimeric form of Jacobsen s catalysts 3, 4. They used the concept of solubility modification by altering the molecular weight of the catalyst so that in a post catalytic work-up procedure the catalyst is precipitated, filtered and used for subsequent catalytic runs. The complexes 3, 4 (0.2 mol % of Co(lll)-salen unit) (Figure 2) were effectively used for HKR of racemic epoxides, e.g., styrene oxide, epichlorohydrin, 1,2-epoxypropane, 1,2-epoxyhexane, 1,2-epoxyoctane, and 1,2-epoxydodecane to achieve corresponding epoxides and 1,2-diols in high optical purity and isolated yields. In this process, once the catalytic reaction is complete the product epoxides were collected by reduced pressure distillation. In 1990, Isono et al. reported the isolation of RK-286C (318) from the culture filtrate and the mycelium extract of Streptomyces sp. RK-286 (303). In nature, this isolate was obtained in its optically active form [a]p -I- 45.3 (c 0.22, EtOAc). Based on similar CD curves, the absolute configuration of this isolate was assigned to be similar to that of (-E)-staurosporine (295) (see Scheme 2.74) (284).
