Oligosaccharide synthesis on polymer support

This chapter discusses recent examples of enzymic oligosaccharide synthesis on polymer supports. Examples include cases where water insoluble acceptor-polymers of improved accessibility are used with soluble enzymes, and cases where water-soluble acceptor-polymers are used with either insoluble or soluble enzymes. The methodology is useful for preparative-scale reactions and for the study of enzymic properties. 

In contrast to oligosaccharide synthesis on polymer supports, the use of polymers as protecting groups in carbohydrate chemistry appears to be a more promising proposition and should be given attention as a convenient alternative to the solution method. 

These results demonstrate that O-glycosyl trichloroacetimidate-based oligosaccharide synthesis on solid support may eventually become a valuable alternative to solution-phase synthesis because useful experience is available for the selection of the polymer support and choice of the linker system and the glycosyl donor. Further standardization of the building blocks and the protective group pattern will finally provide the yields and the anomeric control in order to successfully plan automated syntheses of oligosaccharides also in a combinatorial manner.
