Octams synthesis

In order to replicate, the highly structured and constrained chromatin must be relaxed. Although the nucleosomes are displaced, dissociation of the nucleosome core from the DNA is incomplete, with all the parental histones remaining loosely associated with only one of the parental DNA strands. Synthesis of new histones occurs simultaneously with DNA replication, and nucleosomes containing the newly synthesized histones associate with only one of the new daughter helices. Therefore, the parental histone octamers are conserved, i... [Pg.407]

Nucleosome assembly differs from DNA replication. Histone content of a cell doubles during cell doubling, just as the DNA content does. On the leading strand, the pre-existing histone octamers briefly dissociate from the template and then re-bind to the double helix. Newly made nucleosome core particles associate with DNA on either strand. Thus, the overall process of histone doubling is conservative (the histones stay together during replication), in contrast to DNA synthesis, which is semiconservative. [Pg.231]

Effect of the Structure of Organic Quaternary Ammonium Ions. The tetramethylammonium ion (N C J ), first introduced in zeolite synthesis by Barrer and Denny (30), and Kerr and Kokotailo (21) is effective in forming the cubic octameric silicate anion (Sig02Q°, cubic octamer) (2-16). In the tetramethylammonium silicate aqueous solutions at higher S3.O2 concentrations or cation-to-silica molar ratios (abbreviated to the N/Si ratios), the cubic octamer is singularly formed. [Pg.141]

Kaiser et al. have reported a general entry for the selective synthesis of dimeric macrocycles like cyclostellettamines and for polymeric natural products [41]. It uses the Zincke reaction by which it is possible to control the number of units in a 3-alkylpyridinium polymer. As summarized in Fig. (33), the reaction of the free amine 89 with the Zincke salt 88 gives the dimer 90 (route b) which, after terminal amine deprotection and DNB functionalization at the A-pyridine centre, gives the cyclic dimer, as in the synthesis of cyclostellettamine B. Otherwise, compound 90 furnishes both the protected dimer 91 and the free linear dimer, which, refluxed together in butanol, give the linear tetramer (route c). By the same iterative sequence, the linear octamer was obtained from the tetramer, and from the latter the hexadecamer. [Pg.376]

Hunter, R. F., Turner, C. Chem. Ind., 72 (1957) also reported the synthesis of a cyclic octamer containing six m-bridges and two p-bridges, the remaining four m-positions being blocked with methyl groups... [Pg.46]

Compound 9 is a key intermediate in the synthesis of both linear and branched model octamers containing only free ortho positions. The tetramer 9 can be conveniently depro-tected using tetrabutylammonium fluoride (TBAF)" to afford the first model compound, tetramer 10. [Pg.1639]

Finally, oligonucleotide synthesis of a soluble polymeric support that can be crystallized from the reaction mixture for all the necessary washing steps has been reported [101,102]. This support, based on polyethylene glycol 5000, was loaded at high levels of 3 -0-succinyl nucleosides ( 160 /xmol/g) but allowed only a modest ASWY of 95-97% of octamers. A newer generation of soluble solid supports, based on polymeric assembling of N-acryloylmorpholine, was described as an alternative to the PEG 5000 matrix [103]. [Pg.493]

In 2000 Kohnke and co-workers described the synthesis of meso-octame-thylcalix(6]pyrrole via the conversion of calix[6]furan into dodecaketone, which was then treated with ammonium acetate to obtain calix[6]pyrrole 50 [67]. [Pg.28]

Histones are core proteins which form nucleosomes with DNA, in which an octamer of two copies of each of four different histones H2A, H2B, H3, and H4 are wrapped by DNA. This nucleosome is a basic unit of chromatin. Post-translational modifications of histones, such as acetylation, methylation, and phosphorylation, play an important role in gene regulation [90]. Although the synthesis of histones H2B [91] and H4 [92, 93], which contain no Cys residues, has been reported based on the combination of NCL and a desulfurization strategy by several groups, the synthesis of the Cys residue-containing histone H3 is difficult and has enjoyed limited success [72, 92, 94]. Recently, one-pot synthesis of H3 using peptide hydrazide as a peptide thioester precursor was reported [95]. [Pg.130]
