Nitrilotri methylenephosphonic

Iron(III)-pyrophosphate looks promising as an alternative to iron(III)-carbohydrate preparations for parenteral administration for treatment of anemia.Kinetics of removal of iron from transferrin (tf) by pyrophosphate (pp) were found to be biphasic under certain conditions, with the rapid first phase attributed to the formation of a pp—Fe—tf—CO intermediate.A later study of the kinetics of removal of iron from transferrin employed pyrophosphate and tripodal phosphonates such as nitrilotris(methylenephosphonic acid), N(CH2P03H2)3. For the tripodal ligands there are parallel first-order and saturation pathways, with the latter dominant (contrast the exclusively first-order reaction of ferritin with nitrilotriacetate) for pyrophosphate the paths are roughly equal in importance. The saturation kinetics suggest that tfiFe-phosphonate intermediates play an important role in the kinetics.

NMR spectroscopy was used to follow the binding of alkali metal ions to the Ga(III) complex of the hexadentate ligand formed by linking 3 sets of 3-hydroxy-4( 1 //(-pyridinonc and ethylenoxy chains to tris(car-boxyethoxymethyljethane.1016 31P NMR data revealed that alkali metal ions (Li - Cs) coordinate with nitrilotris(methylenephosphonic acid) (H6L), to form MHL4 and ML5 in solution.1017... 

Nitrilotris(methylenephosphonic acid) (11 n = 3) 322 Ammonium chloride (17.8 g, 0.33 mole), crystalline phosphorous acid (82 g, 1 mole), water (100 ml), and concentrated hydrochloric acid (100 ml) are heated together to the boiling point in a flask fitted with a thermometer, stirrer, reflux condenser, and dropping funnel. 37% aqueous formaldehyde (160 ml, 2 moles, 100% excess) is added dropwise during 1 h and the whole is heated under reflux for a further hour. When the solution is then kept overnight most of the product crystallizes. Collection, washing with aqueous acetone, and drying at 120° affords it (74.9 g, 75%) with m.p. 210-215°. Concentration of the mother-liquor provides a second fraction.

The syntheses of nitrilotris(methylenephosphonic acid) (NTM ), " l-hydroxyethylidene-l,l-diphosphonic acid (HEDP), bis(phosphonyl-methyl) phosphinic acid (PMPA) and tris(phosphonylmethyl) phosphine oxide (PMPO) have been reported. 

One of the most surprising results is the contrast in complexing properties and pH titration curves of PMPO and the published results for nitrilotris(methylenephosphonic acid) N-oxide (ONTMP). Whereas the titration curve for PMPO indicates all 6 protons are removed at pH 10.5, published data for NTMP and ONTMP indicate these materials are mono-protonated at pH 10.5. The values of the pK for monoprotonated NTMP and ONTMP are 12.34 0.14 and 12.05 0.16, respectively, at 25 in the presence of 1.0 M KNO3. Figure 1 shows the variation in P NMR chemical shifts of PMPO upon neutralization with NaOD. The published data for the effect of neutralization upon the chemical shifts of the P NMR signals for ONTMP and NTMP are also given in Fig. 1.
