Dark-field illumination is the most common technique for viewing larval cuticles, particularly for viewing large features such as the mouthparts, ventral denticle belts, and posterior spiracles. A handy substitute for true dark-field illumination is to use low-power objectives with the condenser set in the phase-contrast position for a higher-powered objective (Wieschaus and Niisslein-Volhard 1986). For example, use a lOx objective with the condenser in Ph3 position. Other larval features, e.g., the fine dorsal denticles of the first instar larva, are best observed using phase-contrast optics. [Pg.614]
