The first synthesis of a jasmonoid, namely dihydrojasmone, took place nine years before the structure of jasmone itself was established. This goal was achieved in 1924 by Hermann Staudinger and Leopold Ru icika in the course of their work on pyrethroids. [84] They carried out a Reformatzky reaction between ethyl laevulinate and ethyl 2-bromoheptanoate, and obtained the desired product, although in poor yield, by a Dieckmann cyclisation, followed by hydrolysis and decarbethoxylation (see next page). 

It is well know that copper(II) labile complexes are most widely spread in nature as catalysts of oxidation-reduction reactions and in this sense probably they take second place after iron complexes. On the other hand, however, the copper chelate complexes do not act as catalysts of oxidation, but very often are among the most effective inhibitors of the oxidation reactions. The differences in their catalytic properties may be explained by the fact that all four coordination positions of the central ion are occupied by the chelate ligands. This was demonstrated by Siegel(l) by studying the catalase and peroxidase action of some copper chelate complexes (see scheme 1 in the next page). It was established that the above rate constants sharply decrease in the order I>>II>III of the following scheme and almost reach zero on coordination of the four ligand atoms. 

There is one last mechanistic scenario that we want to cover. It relates to a fully established equilibrium with an intermediate. Remember that by definition the equilibrium constant for a reaction is the quotient of the forward and reverse rate constants. Therefore, the k] / fc ] term in Eq. 7.58 can be replaced by K, which represents the equilibrium constant for the formation of the intermediate. This leads to Eq. 7.60 for such a mechanism (see the Connections highlight on the next page for an example). 

We commonly measure liquid volumes with precision volumetric glassware such as burets, pipets, and volumetric flasks. The National Institute of Standards and Technology in the United States has established specifications for Class A glassware two examples are listed in Table 2.2 on the next page. We may accurately determine the volume of a vessel at one temperature from the mass of a liquid of known density, such as water, that fills the vessel at this temperature. 

Another technique that can be employed to Increase the ease of use of the protocol 1s to establish a format and then keep the format as uniform as possible from one study to the next. In this way, members of the study team will develop familiarity with 1t and will be able to easily find the appropriate page or section of the protocol for resolution of questions. An added benefit to maintaining consistent formats 1s found with the people responsible for generating the documents. The time required to do this necessary part of the operation can be significantly reduced with less chance for errors. 

An antennal-specific aldehyde oxidase (AOX) of M. sexta (MsexAOX) was the next identified pheromone-degrading enzyme (Rybczynski el al., 1989). The activity of MsexAOX was visualized on non-denaturing PAGE, and was shown to be antennal specific but present in sensilla of both male and female antennae. MsexAOX was observed as a dimer with a combined estimated molecular mass of 295 kDa. M. sexta uses a multicomponent pheromone consisting exclusively of aldehydes including bombykal (Starratt el al., 1979 Tumlinson el al., 1989, 1994) MsexAOX was shown to degrade bombykal to its carboxylic acid. Both TLC and spectrophotometric assays were established and a variety of substrates and inhibitors were characterized. Making adjustments for the concentrations and volumes within a sensillum lumen, the in vivo half-life of pheromone was estimated at 0.6 msec in the presence of this enzyme (Rybczynski el al., 1989).
