Negative staining methods

Zinc stain is a negative staining method. This is because the reagent has no effect on proteins but on the background only. This detection method is used in qualitative analysis only, and no destaining protocol is required prior to MS analysis. 

Copper stain— This detection method cannot be used in quantitative analysis because it is a negative staining method. Like the zinc labeling method, copper stain does not require any additional dye removal. However, this technique is compatible with SDS-PAGE only, and therefore, it is not suitable for native separations.

Measurements show some variation depending upon the staining solution used and the method of application. In dried and fixed smears, the cell wall and slime layer do not stain with weakly staining dyes such as methylene blue but do stain with the intensely staining pararosaniline, new fuchsin, crystal violet, and methyl violet. The great majority of bacteria have been measured in fixed and stained preparations. In some instances dried, negatively stained smears have been used. Therefore, the method employed should be specified when measurements of bacteria are reported otherwise the results will be of doubtful v alue. 

A method using the combination of zinc and imidazole produces similar, negatively stained SDS-PAGE gels.56 Zinc imidazolate forms precipitates in gels except at the sites where precipitation is inhibited by SDS-protein bands.57 The resultant gels are opaque white with clear regions at the sites of the protein bands. They too are best viewed with the gel on a black surface. 

As a negative control, the crossreactivities of the secondary antibodies may be checked by exchanging them in single-staining methods. 

We then investigated the formation of hybrid molecular assemblies in combinations of anionic peptide lipid 9 with cage-type hosts 7 and 8 after a previous method [44], Lamella-type aggregates are observed for a mixture of host 7 and lipid 9 at a 1 20 molar ratio in the dispersion state by negative staining electron microscopy. Grids already negatively stained in UA can be rinsed with distilled water, then phosphate buffer and then incubated with antiserum to achieve decoration, followed by re-staining with UA (2). However, personal experience has shown that this is more often than not unsuccessful and it is better to prepare a fresh grid rather than spend time on an unreliable method. 

Autoradiography and staining methods are based on detection of cytoplasmic DNA (in the former case replicating cytoplasmic DNA). For this reason there will always be a low background caused by mitochondrial DNA and samples to be tested should be compared with positive and negative controls.
