Negative staining methods

Zinc stain is a negative staining method. This is because the reagent has no effect on proteins but on the background only. This detection method is used in qualitative analysis only, and no destaining protocol is required prior to MS analysis. [Pg.73]

Copper stain— This detection method cannot be used in quantitative analysis because it is a negative staining method. Like the zinc labeling method, copper stain does not require any additional dye removal. Measurements show some variation depending upon the staining solution used and the method of application. In dried and fixed smears, the cell wall and slime layer do not stain with weakly staining dyes such as methylene blue but do stain with the intensely staining pararosaniline, new fuchsin, crystal violet, and methyl violet.
