Dopamine, a vasodilator, has been widely used for treatment of acute circulatory failure. However, since dopamine is rapidly metabolized when administered orally, its use has been limited to intravenous infusion. Murata et al., studied the bioavailability and the pharmacokinetics of orally administered dopamine (DA). The oral administration of DA to dogs resulted in an absolute bioavailability of approximately 3%. To minimize the extensive first-pass metabolism of DA, a dopamine prodrug, V-(/V-acetyl-l-mcLhionyl)-o,o-bis(cLhoxycarbonyl)dopamine (TA-870), was synthesized [28] (Fig. 6). The homoaromatic interaction in other bridged annulenes has also been examined. The dications of several bridged annulenes were prepared and also studied theoretically and by NMR spectroscopy (Mullen et al., 1987 Wallraff et al., 1988). Once again homoaromatic interactions were deemed to be most important in determining the properties of these systems. Another cationic polycyclic potential homoaromatic system was investigated by Murata and Nakasuji (1980). In situ generation of these cation-radicals has been used in their identification as biological metabolites. A valid alternative was to synthesize the cation-radical as a solid salt, isolate it, and investigate its reactions with various nucleophiles including DNA. From 1985, she was an assistant lecturer at Nara University of Education. She joined the group of Professor Barry M. Trost as a visiting researcher at Stanford University (USA) in 1987-88.
