The elegant genetic studies by the group of Charles Yanofsky at Stanford University, conducted before the crystal structure was known, confirm this mechanism. The side chain of Ala 77, which is in the loop region of the helix-turn-helix motif, faces the cavity where tryptophan binds. When this side chain is replaced by the bulkier side chain of Val, the mutant repressor does not require tryptophan to be able to bind specifically to the operator DNA. The presence of a bulkier valine side chain at position 77 maintains the heads in an active conformation even in the absence of bound tryptophan. The crystal structure of this mutant repressor, in the absence of tryptophan, is basically the same as that of the wild-type repressor with tryptophan. This is an excellent example of how ligand-induced conformational changes can be mimicked by amino acid substitutions in the protein. [Pg.143]

Visit htq) //www-portfolio.stanford.edu/100369. This is Stanford University general chemical safety information from their Portfolio Information System. Also included are storage groups for compatible segregation of chemicals. Chemicals are searchable by scrolling to the end of the section titled Chemical Safety and entering the chemical name. Then select that chemical or other chemical with that chemical root as part of its name. [Pg.183]

The Stanford Linear Accelerator Center, administered by Stanford University, was founded in 1962 as a center for experimental particle physics, but it took until 1966 for its first linear accelerator to be completed. The Stanford Synchrotron Radiation Laboratoiy, built a decade later, became part of SLAC in 1992. Unlike many of other national laboratories that greatly expanded their mission through the years, SLAC always remained a national basic energy research laboratoiy. [Pg.818]
