Quantification of known analytes in PK and distribution studies makes different demands on the analytical procedure than detection of unknown compounds in biotransformation experiments or identification of unknown and postulated molecules in toxicological screening. For example, requirements for quantitative analysis of fixed analytes with optimum sensitivity and selectivity differ from those for qualitative detection of intact molecular weight or diagnostic MS/MS fragments. Selectivity of sample preparation and applicability of diverse scan modes represent relevant critical issues. The following sections address this context. 

The biomedical and medical sciences have increasingly demonstrated that human health and disease are strictly related to molecular processes (Engel, 1977). Each change at the molecular or cellular level is reflected in an alteration of a certain compound in a biological matrix such as tissue, serum, or urine (Spiro, 1975). Consequently, the exact quantitative determination of these components is of the highest diagnostic value (Boutwell, 1975). 

IPG-DALT can be used for the separation of up to 5000 components in a protein mixture, and has now been incorporated as a qualitative tool for diagnostic analysis. It is not generally used for quantitation of proteins in a mixture, however, because loss of low molecular weight components often occurs during the SDS equilibration and transfer steps. Our studies have demonstrated that a molecular luminescence spectrometer with IDA multichannel detector is a powerful quantitative, fundamental, and diagnostic tool. For molecular fluorescence it allows survey spectra to be obtained quickly.
