Migration Alternative test

By way of derogation from condition (a), the comparative test may be dispensed with if there is conclusive evidence based on experimental scientific results that the values obtained in the alternative test are equal to or higher than those obtained in the migration test.

Alternatively, one strip (or, better, half of a strip cut lengthwise) is stained and used as a test for a second strip which is cut into appropriate lengths and eluted. There is, however, no certainty as to the limits of the zones of the protein fractions, for the factors involved in application and migration of protein samples are capable of destroying the parallelism of the two strips and of thus falsifying the results. Nevertheless, this method may be used and is even indispensable for the analysis of lipo-and glycoproteins with reference to the proteins themselves.

An important conclusion from these findings was that even though this analytical method works in principle with olive oil as a food simulant, the migration test using olive oil or another fat simulant can provide false-negative results. Therefore, the method should only be applied in the case of short exposure periods with olive oil. If the method is carried out with olive oil, a recovery check with spiked olive oil applying the same time/temperature migration test conditions is necessary. In the case that such a recovery check indicates loss" of HMDA and/or EDA, then alternatively 95 % ethanol or iso-octane should be used as substitute fatty food simulants. 

Alternatively, if the standing waves are set up vertically using transducers on either side of a small vertical vessel (say a test tube), then any material which migrates and gathers at the nodes will be held static when the vessel is removed in a vertical direction from the ultrasonic field. Essentially the particles are physically being collected by the base of the vessel as it reaches them. This device is known as the Wavecomb [81].
