In a recent paper by Salimbeni et al. [2], a novel series of such All antagonists has been presented on the basis of a comparative analysis of theoretical distributions of the electrostatic potential (inactive compounds and overlay studies, employing a computational model of an All active conformation, it was found that the compound named LR-B/081 [3, 4] (C3oH30N603S), i.e. 2-[(6-butyl-2-methyl-4-oxo-5- 4-[2-(lH-tetrazol-5-yl)phenyl] benzyl -3H-pyrimidin-3-yl)methyl]-3-thiophenecarboxylate (Scheme 1), was one of the most potent in the series, and was selected as a candidate for further studies.

Irradiation of compound 357 (R = Me) in methanol and ethanol at 300 nm gave ethyl 2-methyl-4-oxo-4//-pyrimido[2,l-a]isoquinoline-3-car-boxylate (371) and 3-alkoxy-2-methyl 4//-pyrimido[2,l-a]isoquinoline-4-ones (369, R = Me, R1 = Me, Et) along with two imidazo[2,l-a]isoquino-lines (368, R = Me and 372) and a ring-opened product (92AJC1811). Few completely assigned 13C spectra are available at the time of writing, but it is instructive to compare the chemical shifts for pyrrolo[2,3-(/]pyrimidine (8) (75JA4627) with those of its 6-amino-2-methyl-4-oxo derivative (12 Scheme 2) (78JOC3937).
