Lobe dissection

The collected oocytes need to be individually isolated from the connective ovarian tissue and the layer of follicular cells that surround each oocyte (Goldin, 1992). For a detailed description, see Yao et al. (2000). Briefly, the excised ovarian lobe is washed in MBM and dissected using sterile forceps into smaller sections containing approximately 5-10 oocytes each. The sectioned clumps of oocytes are treated with MBM supplemented with 2 mg/mL type I collagenase (Worthington, Lakewood, NJ) for 2 h at room temperature with constant shaking to separate the oocytes from the follicular cell layer. The oocytes are subsequently washed five times with a BSA solution (bovine serum albumin, 1 mg/mL in MBM), followed immediately by five wash cycles in MBM. [Pg.329]

Most pulmonary tumor colonies in mice are in the pleura. Therefore, pigmented colonies (such as melanomas) can be counted without lung dissection. However, it may be more facile, especially when there are extensive pulmonary metastases, to dissect the lungs into the individual four or five lobes which are then enumerated individually using a dissecting microscope. [Pg.222]

Add glycerol PBS (1 1) to the tube and immediately remove the samples with a cut tip (while they are floating) and place onto a slide. Dissect out the optic lobe with needles and mount the slide with only the eye disc, or retina, on it (see Note 21). Alternatively the slides with imaginal discs can be mounted as previously described for embryos (see Staining and Mounting on Slides ). For dissecting the discs free from the body wall, tear the larva in half and invert the body wall. For dissecting the cluster of dorsal discs (wing, haltere, and second leg disc), pinch the larvae about a third of the way posterior and rip the dorsal anterior side of the larvae a little further anterior. The dorsal cluster is located about a quarter of the way posterior in a dorsolateral position, and often pops out of the resultant tear it must then be teased loose from the trachea. The other leg discs are found on the ventral side of the ventral brain. The eye-antennal discs are stretched between the surface of the optic lobes of the brain and mouth parts. The discs appear slightly more transparent than the white-yellow fat body and can be easily identified with a little practice. [Pg.167]
