Liver defined

Cirrhosis The accumulation of fibrosis in the liver defined by the presence of bridging fibrosis and regenerative nodules. 

Today, ISOpg of peginterferon-a2a once weekly for 48 weeks is recommended in patients with compensated liver disease. However, treatment schedules mat were chosen in clinical trials vary between 24 weeks to 2 years and optimal treatment durations and dose remain to be defined. 

A knowledge of physiology and pharmacokinetics is needed (Fanis et al. 1993 Monteiro and Furness 2001). Levels of mercuiy normally vary among internal tissues, and the time to equilibrate within each tissue varies. For example, blood mercury levels normally reflect veiy recent exposure, while brain and liver levels reflect longer-term exposure. Tissue-specific mechanisms of detoxification and seqnestration, among other processes, must be understood to define the bioactive moiety in observed tissue bmdens before a clear expression of toxicity can be derived (Woodetal. 1997).

Hematopoiesis is defined as the development and maturation of blood cells and their precursors. In utero, hematopoiesis may occur in the liver, spleen, and bone marrow. However, after birth, it occurs exclusively in the bone marrow. All blood cells are generated from a common hematopoietic precursor, or stem cell. These stem cells are self-renewing and pluripotent and thus are able to commit to any one of the different lines of maturation that give rise to platelet-producing megakaryocytes, lymphoid, erythroid, and myeloid cells. The myeloid cell line produces monocytes, basophils, neutrophils, and eosinophils, whereas the lymphoid stem cell differentiates to form circulating B and T lymphocytes. In contrast to the ordered development of normal cells, the development of leukemia seems to represent an arrest in differentiation at an early phase in the continuum of stem cell to mature cell.1... 

ALAD, an enzyme occurring early in the heme pathway, is also considered a sensitive indicator of lead effect (Hemberg et al. 1970 Morris et al. 1988 Somashekaraiah et al. 1990 Tola et al. 1973). Because there is no well-defined blood lead threshold at which inhibition of ALAD does not occur, it allows measurement of the effect on the general population at environmental lead levels and does not require high exposure levels as with occupational workers (Hemberg et al. 1970). However, ALAD activity may also be decreased with other diseases or conditions such as porphyria, liver cirrhosis, and alcoholism (Somashekaraiah et al. 1990).
