Liquid chromatography advantages

Capillary electrophoresis is not used as much as liquid chromatography. Advantages of electrophoresis relative to chromatography include (1) higher resolution, (2) low waste production, and (3) generally simpler equipment. Drawbacks of electrophoresis include (1) higher limits of detection, (2) run-to-run irreproducibility of migration times, (3) insolubility of some analytes in common electrolyte solutions, and (4) inability to scale up to a preparative separation. 

For most samples liquid-solid chromatography does not offer any special advantages over liquid-liquid chromatography (LLC). One exception is for the analysis of isomers, where LLC excels. Figure 12.32 shows a typical LSC separation of two amphetamines on a silica column using an 80 20 mixture of methylene chloride and methanol containing 1% NH4OH as a mobile phase. Nonpolar stationary phases, such as charcoal-based absorbents, also may be used.

In this chapter, the reader has been introduced to the analytical advantages to be gained by linking high performance liquid chromatography to mass spectrometry with particular regard to the limitations of the two techniques when they are used independently. 

The resolution of enantiomers by liquid chromatography using chiral stationary phases is based on the formation of reversible diastereomeric complexes of different stability between the sample and stationary phase. Since the formation of the complexes is strongly dependent on the structure of the sample, there are no universal chiral stationary phases. The specific advantages of TLC for enantiomeric separations result from its low cost, convenience and speed (10,97,98). The main limitation, particularly with respect to column liquid chromatography, is the small number of phases currently available.
