The title compounds LTB4, 21 (Z) and 22 (Z), have been synthesized23 by stereoselective reduction with deuterium gas of a 1 1 mixture of the suitable diacetylenic precursors 23 and 24 using Lindlar catalyst or palladium on barium sulphate catalyst (equation 10). Pygeum africanum, an extract from the bark of the African prune tree, has been used in Europe since 1969 to treat men with mild to moderate symptoms of benign prostatic hyperplasia. The mechanism of action of P. africanum is not known. In animal models, P. africanum modulates bladder contractility, has anti-inflammatory activity, decreases production of leukotrienes and other 5-lipoxygenase metabolites, inhibits fibroblast production, affects adrenal androgens, and restores the secretory activity of prostate epithelium. [Pg.513]

Liquid C h rom atography/M ass Spectrometry. Increased use of liquid chromatography/mass spectrometry (lc/ms) for structural identification and trace analysis has become apparent. Thermospray lc/ms has been used to identify by-products in phenyl isocyanate precolumn derivatization reactions (74). Five compounds resulting from the reaction of phenylisocyanate and the reaction medium were identified two from a reaction between phenyl isocyanate and methanol, two from the reaction between phenyl isocyanate and water, and one from the polymerization of phenyl isocyanate. There were also two reports of derivatization to enhance either the response or structural information from thermospray lc/ms for linoleic acid lipoxygenase metabolites (75) and for cortisol (76). [Pg.246]

Lipoxygenase metabolites of arachidonic acid such as 12-hydroxyeicosa-tetraenoic acid (12-HETE) have been shown to be involved in the release of neurotiansmitters and other cellular functions (92-94). Basal generation of 12-HETE has been reported to be about 3.7-fold greater in SHR than in WKY platelets (95,96). Arachidonic acid has been shown to stimulate platelets by activating protein kinase C, by a prostaglandin-independent mechanism (97). It is possible that the lipoxygenase metabolites of arachidonic acid may also be involved in altered reactivity in hypertension. [Pg.451]

Platelets in hypercholesterolemic patients are more susceptible to aggregation than normal subjects [72]. Aggregation is mediated by several mediators, including cyclooxygenase and lipoxygenase metabolites of arachidonic acid [73].
