Lipoxygenase cloning

Soybean lipoxygenase-1 also acts as a 15-lipoxygenase for AA, but the amino acid identity between human 15-lipoxygenase and the soybean enzyme is only 22% [77]. cDNA encoding 15-lipoxygenase cloned from human bronchus cDNA library [78] is identical with that from human reticulocyte cDNA library [77]. Thus the presence of two forms of 15-lipoxygenase in a human leucocyte preparation [82] awaits further investigation.

The cloning, characterisation and expression of many lipoxygenase (TOX) [17] and hydroperoxide lyase (HPL) [18] genes has led researchers to propose new processes for the production of green note flavours. HPT specifically produces the highly demanded compound ds-3-hexenal from the 13-hydroperoxide of linolenic acid and hexanal from the hydroperoxide of linoleic acid, both of which are formed by TOXs (Scheme 26.2).

Another example is the production of green note compounds using lipoxygenase pathway enzymes. Lipoxygenase and hydroperoxide lyase are the determinant enzymes for the conversion of fatty acids into natural food flavor components. However, these compounds present in natural sources at very low levels and because of their instable nature it has been difficult to purify these compounds. As a result, considerable efforts have been made to clone these two enzymes for commercial uses in the production of natural flavor components.
