B7-H4 are upregulated on the surface of DCs upon contact with Tregs [15, 16]. Although for both molecules the counter-receptor is not known, it is most likely that immunosuppressive signals will be conveyed. Functional data concerning B7-H3 and B7-H4 are still limited, since they have been described rather recently. Nevertheless, similar to other members of the B7-H family, B7-H3 has clear immunosuppressive functions, as this molecule has been shown to mediate the reduction of autoimmune EAE and addition of recombinant B7-H3 molecules to mixed leukocyte reactions suppresses T-cell proliferation in vitro [17-20].

When these same gangliosides were added to mixed leukocyte cultures (MLC), all caused some inhibition at 12.6 nanomoles and none inhibited when 1.6 nanomoles was added (Table V). It is preferable but not essential to screen PBMCs from different donors to identify a donor whose cells replicate HIV efficiently and do not carry the A 32 C025 mutation. Alternatively, leukocyte enriched buffy coats may be purchased from blood transfusion centers. Mixing PBMCs from two donors results in a mixed lymphocyte reaction which can increase cellular activation and hence HIV replication. 

The reaction mixture was prepared by mixing 800 fiL of substrate solution containing 0.01 M p-nitrocatechol sulfate (dipotassium salt), 1.71 M NaCl, and 0.5 M acetate buffer (pH 5.0) with 200 fiL of either leukocyte and platelet lysate or saliva. Leukocyte lysates adjusted to equal protein content were maintained at 4 in buffer. At indicated times aliquots were removed and enzyme activity determined. The size of the aliquot was constant for each subject. The assay mixture contained 10 Ji liters IM tris buffer, pH 8.0, 50 jl liters 0 02 M MgCl2 and 50 jl liters lO M PRPP (mixed just before assay 110 JL liters of the mixture used per tube), 50 [x liters of 1 50 lysate.
