Knock-down efficiency

This same strategy can be attempted at the mRNA level to inhibit the translation of individual mRNAs. These experiments are almost never 100% efficient hence, the problem becomes how much neutralization is necessary to elicit the desired effect. If the antisense oligonucleotide is targeted toward the mRNA for a particular transcription factor or splice form of a transcription factor, then it may be possible to knock down or reduce the expression of that particular protein. The result of such a knockdown will be a new balance of transcription factor subunits in the cell, probably resulting in alteration of the relative amounts of particular homo- and heterodimers. [Pg.469]

The primary application of LNA has been in antisense therapy due to the aforementioned properties. It has been used to knock down human protein kinase C-a (PKC-ot) with greater efficiency than the corresponding phosphor-othioate ODN, as an inhibitor of human telomerase, to block the synthesis of RNA polymerase II and to inhibit human vascular smooth muscle cell growth. LNA-modified oligonucleotides have also been used in RNA interference (RNAi). ... [Pg.720]

Transfection is conducted to introduce nucleic add (NA) con-stmcts, such as DNA or RNA, to either overexpress or knock down the produrtion of a specific protein in the cell. This is often used in molecular biology, and is the focus of ne therapy. As experted, this is complicated in vivo, and the effective delivery of NA constmcts for gene therapy treatment is hampered by the rapid degradation of NA in circulation. It is also challenging to target tissue spedlically and also the low efficiency of intracellular delivery and transport to the nudeus further hampers the efficiency. Therefore it is necessary to package the NA to shield it and increase the efBdenty of transfertion. Often cationic liposomes or cationic dendrimers... [Pg.163]
