Jigsaw

Normal ions (M+, Fj+,. .., F +) in a spectrum can provide a molecular structure for substance M if the fragments can be theoretically reassembled. The problem is rather like deducing an original jigsaw puzzle by putting the pieces together correctly. For most molecules containing more than a few atoms, this reassembly exercise is difficult and often problematic. 

To carry out the reassembly exercise, connectivities between the ions must be determined. For example, does the ion Fj+ come from M+ or F/ or F2+ This process can be likened to completing a jigsaw by finding where and how the pieces fit together. 

Analysis of Data. A veteran practitioner of chemical market research likened this step to the assembly of a jigsaw puzzle. There are many pieces of unequal size and importance that must be put together to make a picture understandable to everyone. Call reports, secondary data inputs, experience, and judgment are the tools used by the market researcher to analyze the data, reach conclusions, make recommendations, and write the report. 

Five double-bond equivalents can be recognised from the shift values (four for the benzene ring and one for the carbonyl group). The sixth double-bond equivalent implied by the molecular formula belongs to another ring, so that the following pieces can be drawn for the molecular jigsaw puzzle ... 

A completed jigsaw puzzle can be disassembled by a sweep of the hand, but shaking a box containing Jigsaw puzzle pieces never generates an assembled puzzle. 

Each of these processes has a spontaneous direction, in which matter becomes more spread out or dispersed. Another way to describe these processes states that each reduces the constraints on the objects. The marbles in a bag, the boards of a fence, and the interlocking pieces of a jigsaw puzzle are restricted, or constrained, in their positions. In the case of a total unknown it is a case of the more data, the better. Solving this sort of problem is like doing a jigsaw puzzle. You piece together information from a variety of sources to come up with a feasible structure. You then test that structure with more experiments to ensure you get a consistent answer. As a minimum you should consider COSY, HSQC, HMBC, 1D 13C. Don t forget-NMR is not the only technique so look at mass spectrometry (accurate mass in particular) and IR to help. 

Thus, the approaches, models, hypotheses, theories, experiments and computer simulations presented in the various chapters, as well as the speculations and lively controversies, have not yet led to a clear picture of the origins of life. While the individual research results discussed above provide important contributions, and individual pieces of the jigsaw puzzle, vital parts of the complete picture are still missing the process of life s emergence from inanimate precursors can still not be described satisfactorily. 

There is no relevance to the order of the items in the list. In fact there is no agreed order in which to evaluate the characteristics. Method validation is like putting together a jigsaw puzzle . The more pieces that are in place, the clearer the picture becomes. The difference is that in validation all of the pieces are not always required. Table 4.6 shows when a study of the parameter is required for four different situations. However, it is important that validation is a planned activity or otherwise it can become very labour-intensive and inefficient. Some familiarity with the statistical terms introduced in Chapter 6 is essential before starting out to plan method validation.

The power of LC-MS over MS as a single technique is the ability to separate out the components of a mixture and then provide MS data on each. The use of accurate mass LC-MS is increasing as it enables the molecular formula of the analyte to be determined as an additional piece of information in the structural elucidation jigsaw. 

Before structural work can begin, pure substances must be separated from the complex mixtures in which they occur in cells and tissues.413-24 Often, a substance must be isolated from a tissue in which it is present in a very low concentration.
