DiaZepin Nucleosides. Four naturally occurring dia2epin nucleosides, coformycin (58), 2 -deoxycoformycin (59), adechlorin or 2 -chloro-2 -deoxycoformycin (60), and adecypenol (61), have been isolated (1—4,174,175). The biosynthesis of (59) and (60) have been reported to proceed from adenosine and C-1 of D-ribose (30,176,177). They are strong inhibitors of adenosine deaminase and AMP deaminase (178). Compound (58) protects adenosine and formycin (12) from deamination by adenosine deaminase. Advanced hairy cell leukemia has shown rapid response to (59) with or without a-or P-interferon treatment (179—187). In addition, (59) affects interleukin-2 production, receptor expression on human T-ceUs, DNA repair synthesis, immunosuppression, natural killer cell activity, and cytokine production (188—194). [Pg.124]

Studies have actually revealed two type I interferon receptor polypeptides. Sequence data from cloning studies place both in the class II cytokine receptor family. Both are transmembrane N-linked glycoproteins. Studies using isolated forms of each show that one polypeptide (called the a/p receptor) is capable of binding all type I interferons. The other one (the ap receptor) is specific for IFN-a-B (a specific member of the IFN-a family). Both receptors are present on most cell types. [Pg.215]

Drug interactions No drug interaction studies have been conducted with Avonex. Other interferons have been found to reduce cytochrome P-450-mediated drug metabolism. Hepatic microsomes isolated from Avonex-treated rhesus monkeys showed no influence on hepatic P-450 enzyme metabolism activity. [Pg.195]

The interferon is produced within the E. coli cells, which must first be separated from the culture media by using centrifugation. The isolated cells are then solubilized by cell disruption, after which the fraction containing interferon is concentrated by salting-out. Two subsequent procedures using immunoaffinity and cation-exchange chromatography raise the purity of the interferon 1000-fold. [Pg.236]

Dr. Alick Isaacs and Dr. Jean Lindenmann discovered interferon in 1957 in Great Britain. In an experiment, they mixed five viruses with egg membranes and unexpectedly found that the viruses did not grow. They did not know what caused this observation and simply indicated something interfered with the virus, so the term interferon was coined. They eventually identified the polypeptide and were able to isolate it from various cells, including those taken from calves, monkeys, and humans. [Pg.244]
