Insertions, gene

Vector (i) Plasmid or virus used in genetie engineering to insert genes into a eell. (ii) Agent, usually an inseet or other animal, able to earry pathogens from one host to another. [Pg.628]

An additional consideration that may influence the protocol used is the desired duration of subsequent expression of the gene product. In most cases of genetic disease, long-term expression of the inserted gene would be required. In other instances (e.g. some forms of cancer therapy or the use of gene therapy to deliver a DNA-based vaccine), short-term expression of the gene introduced would be sufficient/desirable. [Pg.423]

More recently, various modifications have been introduced to this basic retroviral system. The inclusion of the 5 end of the gag gene is shown to enhance levels of vector production by up to 200-fold. Additionally, specific promoters have been introduced in order to attempt to control expression of the inserted gene. Most work has focused upon the use of tissue-specific promoters in an effort to limit expression of the desired gene to a specific tissue type. [Pg.426]

The aspect to which far more attention has been directed is the nature of the inserted gene(s) and promoters in rDNA products. Again, the toxicologist should ask how well the nucleotide sequence is known, whether there is only one reading frame, and how any introns are handled. Again, toxicity-type testing would appear to be an inefficient and expensive way to study molecular biology and biochemistry. [Pg.434]

These DNA viruses can invade cells that are not dividing, providing an advantage over retroviruses. Disadvantages include the fact that adenoviruses do not insert their DNA into the DNA sequence of the host, so the inserted gene is eventually lost. Repeated administrations are therefore necessary. Adenoviruses, although modified, may still provoke an immune reaction. [Pg.351]

The cells are first made competent for transformation by treatment with calcium chloride using a standard procedure (Appelbaum and Shatzman, 1999). Recombinants containing the inserted gene can be conveniently screened by PCR, using vector-specific and gene-specific primers. [Pg.3]

When scientists first successfully inserted genes into microbes and plants, it was the potential to exploit this science commercially that was sold to, and excited, politicians... [Pg.216]

Linearize 20 pg of the plasmid by digestion with Pvul (see Note 6 if the inserted gene includes a Pvul site). Precipitate the digested DNA with ethanol and dissolve in 50 pL sterile deionized H20. [Pg.434]
