Induction of oxidative stress

Genotoxic modes of action involve genetic alterations caused by the substance interacting directly with DNA to result in a change in the primary sequence of DNA. A substance can also cause genetic alterations indirectly following interaction with other cellular processes (e.g., secondary to the induction of oxidative stress). 

Chronic administration of peroxisome proliferators to rodents results in sustained oxidative stress due to overproduction of peroxisomal hydrogen peroxide. The induction of peroxisomal fatty acid P-oxidation by cinnamyl anthranilate in vivo under bioassay conditions (Lake et al, 1997) supports this h othesis. Other data on the induction of oxidative stress are not available for cinnamyl anthranilate.

Without a mechanism for its excretion, iron accumulates in vital organs (Pietrangelo, 2002). Because the liver binds both circulating nontransferrin and transferrin-bound iron, the liver is at particular risk for iron overload. Excess iron causes damage to hepatocytes primarily through induction of oxidative stress (Parkilla et al., 2001).

Interactions between fragments of a-synuclein and Afi peptide promote the aggregation of a-synuclein in vivo. In addition under pathlogical condition, interactions between Afi and a-synuclein may initiate the formation of toxic oligomers and nanopores that increase intracellular calcium leading to induction of oxidative stress, leakage of lysosomal membranes, and mitochondrial dysfunction (Crews et al., 2009).
