Incubation test

An important DMPK property of a NCE is oral bioavailability (F) of the compound in various pre-clinical species.3 The oral bioavailability of a compound is dependent on several factors including intestinal permeability (estimated by the Caco-2 assay) and hepatic clearance (estimated with an in vitro metabolic stability assay).3 30 The metabolic stability assay is typically performed by incubating test compounds in liver microsomes or hepatocytes. The results can provide estimates of in vivo stability in terms of metabolic liabilities.3 8 59 62 Several authors described this assay as an important tool for the rapid assessment of the DMPK properties of NCEs.3 6 8111819 26 44 59 62-65... [Pg.209]

In addition to standard incubations, tests were performed. Dose-response curves were made with dilution series of whole mesocosm samples in artificial sea water. The effect of the incubation temperature (15°C) was also compared with a series at ambient temperature (4°C, all sample handling and incubation in cold room). Effects of light conditions were tested by incubations in the dark (wrapped in aluminium foil), 7 and at 40 gmol photons m 2 s-1. [Pg.192]

Microbiological checks are usually centred on the bright beer tank, the filler and seamer units, and particularly on the canned beer emerging from the pasteurizer. The latter is membrane-filtered and the membranes are incubated on nutrient medium there should be no colonies after 3-4 days at 25-30°C (77-86 F). Some cans are held for 14 days at 30°C (86°F) before being subjected to the membrane-filtration and incubation tests. Again there should be no development of colonies. [Pg.350]

The fjJADP(H)-content of the total protoplasts was 20-25 nmol mg Chi in all conditions of incubation tested. Most of this (70-80 %) was found in the chloroplasts except when Gald was present, then the chloroplast pool was only 45 % of the total NADP(H)-pool. Almost all of the extrachloroplastic NADP(H) was cytosolic, less than 2% was associated with the mitochondria. [Pg.2783]

Solutions for incubation tests were obtained by mixing bovine synovial fluid (BSF) with labeled BSA, labeled AGP and labeled AlAT to concentrations of 12.5 mg/ml for BSA and of 0.3 mg/ml for the glycoproteins. BSF was removed from the knee Joint of cows not older than 18 months at the HinwiI slaughterhouse in Switzerland. All samples were clear, yellow, particle-free liquids and were stabilized with protease inhibitor (Sigma-Aldrich, Switzerland) and 0.1% sodium azide. [Pg.406]
