Incubation conditions, selection

Incubation conditions shall be suitable for recovery of bioburden and environmental isolates at 20 to 35°C. Microbiology department shall provide a rationale for the selection of their specihc incubation conditions according to the trend analysis of area environmental control and presterilization bioburden isolates. The temperature chosen shall be based upon its ability to recover microorganisms normally found environmentally or in the product bioburden. This same panel of microorganisms shall be used in growth testing the media-hlled containers. [Pg.907]

An extensive study of the effects of formaldehyde in milk lipase inhibition showed that formaldehyde acts as a competitive inhibitor and, under the proper conditions, selectively inhibits the lipases of raw skim milk (Schwartz et al. 1956A). This study showed that the inhibitory effect of formaldehyde was dependent on such factors as pH, time of addition of the inhibitor, length of the incubation period, concentration... [Pg.230]

C, and then move the filled containers to 20-25°C. The lack of consensus suggests that the selection of incubation conditions is likely a minor concern. The suitability of the conditions employed, whatever they might be is established by the conduct of growth promotion studies (see following section). The one constant in this area is the duration of the incubation period which is almost universally set at 14 days. ... [Pg.135]

C) and incubation time (10-60 min) for Lu-P3 are shown in Fig. 16.6. Radiolabelling yields of greater than 99% were obtained when reactions were carried out for either 10 min at 70°C or 30 min at 55°C. In contrast, 80% yield was obtained at 40°C, even when the reaction was carried out for 60 min. The conditions selected for subsequent labelling studies were incubation for 30 min at 70°C. Similar results were obtained for the radiolabelling of P4 and OCT with Lu. [Pg.283]

The first step before incubations with antibodies is the use of detergents to solubilize the membranes of the cells and tissues. As discussed in Chapter 5, selection of a detergent and its incubation conditions are important. The best penetration of antibodies is found after removal of as much of the cellular membranes as possible. [Pg.100]

The duration of the fish incubation with the chemical prior to irradiation also affects the phototoxicity of a-terthienyl. In the experimental conditions selected, the survival was 100% when the fish were irradiated for 30 min immediately upon contact with the solution, 0% when irradiated after 2 h, and 70% when the irradiation was started 8 h after initial contact. This indicated that a) a minimum incubation period was required for the sensitizer to reach the biological targets leading to lethal damage, and b) depuration took place. However, nothing is known about the relationship between the initial concentration of sensitizer and the magnitude of this phenomenon (757). [Pg.152]

Of the 8 fatty acids tested as giganteum lipoxygenase substrates (delta 9,12-octadecadienoic, delta 9,12,15-octadecatrienoic, delta 6,9,12-octadecatrienoic, delta 11,14-elcosadienoic, delta 8,11,14-eicosatrienoic, delta 5,8,11,14-eicosatetraenoic, delta 5,8,11,14,17-eicosapentaenoic, and delta 4,7,10,13,16,19-docosahexaenoic acids, all-cis), only arachidonlc (C-20 4), eicosapentaenoic (C-20 5) and docosahexaenoic (C-22 6) acids were consistently metabolized (Table 1). It is noteworthy that the C-18 compounds apparently are not suitable substrates for the giganteum enz5nne under the incubation conditions used. If this initial observation is verified by work in progress, this will be the first documented instance of such selective lipoxygenase substrate specificity. [Pg.422]
