In vitro binding

This approach can be generalized to all possible types of experimental data that may be generated. All chemical structures available in public databases or internal to a company typically feature at least the in vitro binding assay data and additionally, the three-dimensional structure of the protein and/or bound ligand. A chemical compound C will therefore be ... [Pg.335]

Among the D2 family of receptors (D2, D3 and D4) the D2 receptor itself seems to be the most important. At a therapeutic concentration, most neuroleptics, except clozapine (and risperidone), should, according to in vitro binding studies, be occupying 50-70% of brain D2 receptors. The picture is similar for D3 receptors but only clozapine (and... [Pg.364]

In vitro binding studies use different cell lines or membrane preparations and generally only yield the apparent dissociation constants for a number of antagonists obtained by comparative displacement of one labelled ligand. Unfortunately few... [Pg.367]

The Fur protein from E. coli was isolated in one step due to its high affinity for metal-chelate columns loaded with zinc. In DNase footprinting experiments, the Fur protein was shown to bind DNA in the promoter region of several iron-regulated genes. The consensus sequence, called the Fur box, is GATAATGATAATCATT ATC. In vitro binding is dependent on the divalent cations Co2+ Mn2+ /s Cd2+ Cu2+ at 150 iM, while Fe2+ seemed to be less active at this concentration, probably due to oxidation to Fe3+ (De Lorenzo et al., 1987). The unspecificity for divalent metals observed in vitro shows that the cells have to select the ions transported carefully and have to balance their active concentrations. In addition, it is a caveat for the experimenter to test a hypothesis on metal-ion specificity not only in vitro, but also in vivo. [Pg.108]

With the introduction of chlorpromazine in 1952, there was a small revolution in psychiatry patients suffering from psychosis were able to be de-institutionalized. Chlorpromazine and other typical antipsychotics (e.g., haloperidol) demonstrate high in vitro binding affinities for the dopamine D2 receptor (D2). Specifically, their... [Pg.370]

It has been noted that cell cultures derived from trout (66, 67) and trout liver microsomes (U2, U7, 68) may have relatively high AHH activity (BP hydroxylase) which sometimes exceeds the activity observed in control rat liver microsomes. The metabolite pattern obtained using trout liver microsomes resembled that produced by MC treated rat liver microsomes (P-Uh8)(6 and Ahokas, Saarni, Nebert and Pelkonen, manuscript in preparation Table IV). Associated with this pattern of BP metabolites was covalent binding to DNA which was three times as high as obtained by using rat liver microsomes. Another species of fish (roach), on the other hand was found to be almost inactive in catalyzing in vitro bind-... [Pg.287]
