Goldenseal is yet another threatened herb in the United States and other parts of the world. Fibroblasts were selected because they are readily cultivated and radio-labeled and are available from skin explants of a variety of species. Autoradiograms of radiolabeled cellular proteins are more diverse and easily analyzable for molecular systematics than alternatives such as silver-stained serum or erythrocyte protein patterns. We have achieved nearly 100% success rates at minimal discomfort and risk to human subjects by establishing cultures from 3 mm punch biopsies from the upper buttock. Local lidocaine anesthesia is used. Samples are collected following informed consent and under an approved human research protocol. Other species are sampled by dart gun8 or while sedated. Skin samples can be collected from various body sites without compromising the 2D electrophoresis metric, which does not rely on quantitative differences in protein expression. To comply with the Convention on International Trade in Endangered Species of Wild Fauna and Flora (CITES), tissues collected from wild and captive exotic animals must be obtained under specific permits issued by the U.S. Fish and Wildlife Service. 

A distinction is made between academic and industrial requests for access. Commercial requests must be submitted to MEEFT at the CITES (Convention on International Trade in Endangered Species of Wild Fauna and Flora) department and possibly also to the Ministry of Trade. The procedures are quite simple for research purposes (small quantities of biological materials). But for commercial/industrial activities and CITES species, there are specific regulations such as the need to provide proof of the existence of a nursery from which plants have been collected there is also a quota.

Labeling to identify destination address and, where appropriate, customs agents at the destination airport is required. If samples are of animal origin, specifically those covered by CITES (Convention on International Trade in Endangered Species), it is essential that clearance is obtained from both the sending and receiving countries relevant authorities. Without this, delays will ensue and more than likely samples will be compromised.

Each year nearly 13,000 animals arrive at North American zoos and aquariums. Of those, about 5000 are born there. The rest, according to the internationai Species information System, are transported by plane, truck, or boat from across town or from remote locations across the world (Davis, 2004). Some 5000 species are protected under an agreement called the Convention on International Trade in Endangered Species of Wild Fauna and Fiona (CITES). With a conservation mission, zoos and aquariums have begun to produce more wild animals than they consume. More than 90% of mammals and 70% of birds on display were born in captivity. 

Such precautions are particularly important in endangered areas. Where there are threats of substance, it is important to carry out regular geodesic measurements along the route and to measure the pipes for possible stretching.
