We investigated the efficiency of NSC expansion on surfaces with EGF-His immobilized in the correct orientation. NSCs were obtained from neurosphere cultures prepared from fetal rat striatum harvested on embryonic day 16. NSCs were cultured for 5 days on EGF-His-immobilized substrates prepared with mixed SAMs of different COOH-thiol contents. Cells adhered and formed network structures at a density that increased with the COOH-thiol content of the surface. As a control, cells were seeded onto surfaces without immobilized EGF-His. This resulted in poor cell adhesion during the entire culture period. In addition, when EGF-His adsorbed to SAMs with 100% COOH-thiol or SAMs with NTA-derivatized COOH that lacked Ni2+ chelation, we observed poor initial cell adhesion, and the cells formed aggregates within 5 days. Interestingly, the substrate used to covalently immobilize EGF-His with the standard carbodiimide chemistry was not a suitable surface for cell adhesion and proliferation. The control experimental results contrasted markedly with results from EGF-His-chelated surfaces. [Pg.181]

Off-line dicarbamate solvent extraction and ICP-MS analysis [317] provided part-per-trillion detection limits Cd (0.2 ppt), Co (0.3 ppt), Cu (3 ppt), Fe (21 ppt), Ni (2 ppt), Pb (0.5 ppt), and Zn (2 ppt). Off-line matrix removal and preconcentration using cellulose-immobilized ethylenediaminetetraacetic acid (EDTA) have also been reported [318]. Transition metals and rare earth elements were preconcentrated and separated from the matrix using on-line ion chromatography with a NTA chelating resin [319]. Isotope-dilution-based concentration measurement has also been used after matrix separation with a Chelex ion-exchange resin [320]. The pH, flow rate, resin volume, elution volume, and time required for isotope equilibration were optimized. A controlled-pore glass immobilized iminodiacetate based automated on-line matrix separation system has also been described [321]. Recoveries for most metals were between 62% and 113%. [Pg.134]

Applications. A biotinylated GOX-based biosensor was developed based on a new electropolymerized material consisting of a pol3rp3uidyl complex of ruthenium(II) functionalized with a pyrrole group [90]. Because histidine, lysine and arginine functions also coordinate Os /Os , biosensors based on co-electrodeposited GOX, HRP, soybean peroxidase (SBP) and laccase with redox Os /Os polymer have been developed [89]. A metal chelate formed by nickel and nitrilotriacetic acid was used to modify a screen-printed electrode surface. The functionalized support allowed stable attachment of acetylcholinesterase and the resulting biosensor was used for sensitive detection of organophosphorus insecticides [91].
