Toxicology hypothesis testing

Testing toxicological perturbances for the analysis of quantitative structure-activity relationships requires a rigorous approach to rationalizing effects. To date, emphasis has been placed on assessing toxicity in terms of basic physico-chemical relationships. However, there has frequently been difficulty in interpretation when data from different laboratories ai e compared, as levels of apparent toxicity often differ. Focusing on validation process of in vitro methods, it is possible to underline some differences between tools for research and ones for toxicological testing. A research model is validated when there are some specific evidences confirming that the information from the model is able to correctly describe the process in the intact animal. Tools for toxicity testing are often used to evaluate safety hypothesis so they can be used without requiring in vivo confirmation. They are validated using a subset of well-known materials and, once validated, systems will be applied to new unknown materials or mixtures in order to evaluate their toxicity and compare their potential with other chemicals. 

M.B. Schenker of Institute of Toxicology, University of California Davis in Davis, California is leading a multidisciplinary study supported by National Cancer Institute (NCI). This study will examine whether environmental asbestos deposits in California are associated with increased rates of mesothelioma. The study will address geological occurrence of asbestos and potential human exposure based on population patterns and known occupational exposure, and epidemiological characteristics of the disease in the state. The project will plan a case-control study to rigorously test the hypothesis that mesothelioma in California is independently associated with environmental asbestos exposure.
