Schallmoser, K., Bartmann, C., Rohde, E., et al. (2007), Human platelet lysate can replace fetal bovine serum for chnical scale expansion of functional MSC, Transfusion. 2007 Aug 47(8), 1436-1446. [Pg.113]

Several acidic NAA have been shown O to Inhibit nlcotlnate phospho-rlbosyl transferase in human platelet lysate by reversible competition with nicotinic acid. The resulting suppression of nicotinamide adenine dl-nucleotlde biosynthesis may Inhibit mucopolysaccharide biosynthesis and reduce the Inflansnatory response. [Pg.209]

In this communication we report on the presence of both PRPP synthetase and HGPRT in human platelet lysates. The activity of these enzymes was also studied in rabbit platelets. Platelets were separated and washed according to Ardlie et al. (5) and suspended in tris-buffered saline pH 7.35 (3). The incorporation of purine bases into the nucleotides of intact platelets was investigated by incubating cell suspensions in tris-buffered saline containing labelled purine bases. The reactions were arrested by the addition of perchloric acid and the total nucleotides in the protein-free supernatant were separated from the purine nucleosides and bases by thin... [Pg.159]

It was found that human as well as rabbit platelet lysates synthesize PRPP from ribose-5-phosphate and ATP. The average specific activities of PRPP synthetase in pooled human and pooled rabbit platelet lysates were 12 and 22 nmoles/mg protein/hr, respectively. [Pg.160]

Both human and rabbit platelets were found to possess, in addition to APRT (2) also HGPRT activity (Table l). The specific HGPRT and APRT activities and the ratio between them are similar to those found in cultured human skin fibroblasts (8), but differ from those in human erythrocyte lysates in which the specific activities of both enzymes are lower, especially that of APRT (9). Human and rabbit platelets were also found to... [Pg.160]
