Human fibrinogen, preparation

This component, present in plasma but not in serum, is of fundamental importance, but as yet the data available concerning it are few. Holm-berg (105) has reported for human fibrinogen, prepared by repeated salting out with ammonium sulfate, sodium chloride solution and phosphate buffer, S20 = 8.5 S, Djo = 1.1 X 10", and M 700,000. The fibrinogen preparations obtained in this laboratory by the low salt, low temperature ethanol precipitation methods discussed in Part VI, have been studied extensively by Oncley (162). This is a dry sponge of human fibrin prepared by elotting a foam of human fibrinogen solution with human thrombin. It is then freeze-dried, cut into shapes and sterilized by dry heat at 130°C for 3 hours. Before use, it is saturated with thrombin solution. Blood coagulation occurs in contact with the thrombin in the interstices of the foam.

Preparation of Washed Red Cells. Freshly drawn human blood, collected into acid citrate dextrose (ACD) anticoagulant, was centrifuged for 15 min at 630 X g, and the plasma and huffy coat were removed. The red cells were washed three times using 3-4 volumes of PBS, centrifuging between each wash at 2000 X g for 10 min.
