Hematoxylin stain

Iron hematoxylin Stain or dye to identify cell nuclei or identifiable characteristics of an organism. [Pg.1569]

A number of staining procedures have been described. Some stains, such as chlorazol black, require fresh specimens and are not widely used. A variety of stains for fecal smears preserved by Schaudinn or PVA fixative have been described, including various hematoxylin stains. The stain most widely used in the United States is the Wheatley trichrome stain, which is the only permanent stain described in this chapter. The trichrome staining procedure uses reagents with a relatively long shelf life and is easy to perform. Note that there are differences in staining times depending on whether the specimen is fixed in Schaudinn or PVA fixative, as penetration is slower in the latter. [Pg.17]

An appropriate dilution of the heparinised BM aspiration is needed for accurate enumeration of the primary CFU-F frequency as indicated in this representative experiment where whole heparinized BM was seeded corresponding to the respective measured BM-MNC number per square centimeters of growth area, cultured for 11 days at 37°C/humidified atmosphere/3% 02/5% C02. Ple6nik (1902) was among the first to apply Weigert s chrome-hematoxylin stain to adrenal glands fixed for a prolonged period in potassium dichromate mixtures. He found that the droplets in the adrenal cortex stained intensely after such mordanting, as did those of the interstitial cells in the testis, but that ordinary fat failed to stain. [Pg.175]
